Penicillin G Procaine is prepared by the reaction of dibenzyleth-ylene diamine with two molecules of penicillin G. It is chemically designated as (2S,5R,6R)-3,3-Dimethyl-7-oxo-6-(2-phenylac-etamido)-4-thia-1-azabicyclo[3.2.0]heptane-2-carboxylic acid compound with N,N'-dibenzylethylenediamine (2:1),tetrahydrate. It occurs as a white, crystalline powder and is very slightly soluble in water and sparingly soluble in alcohol. COMPOSITION Penicillin G Procaine 200,000/IU Dihydrostreptomycin sulphate 250,000/IU Solvents ad 1ml or Penicillin G procaine 200,000/IU Dihydrostreptomycin sulphate 200,000/IU Solvents ad 1ml Experimental: Reagents and apparatus: Penicillin G potassium (PG) and Penicillin G Procaine (PGp) standards were purchased from Wako Pure Chem. (Osaka, Japan) and Nippon Zenyaku Kyogyo (Koriyama, Japan), respectively. Ethanol (purity 99.5%) and distilled water were of HPLC grade (Wako). Stock standard solutions of PG and PGp were prepared by dissolving each of the compounds in water to a concentration of 100 µg/mL. Working mixed standard solutions of these compounds were prepared by diluting the stock solutions with water. A handheld ultrasonic-homogenizer (model HOM-100, 2 mm i.d. probe, Iwaki Glass, Funabashi, Japan), a micro-centrifuge (Biofuge fresco, Kendo Lab.
